Protein-Protein Networks
Description:The aim of this Think Tank is:
1. A critical evaluation of the (potential) merit and pitfalls of large-scale (tandem) affinity tag purification in conjunction with proteomics in deciphering protein interaction networks. The several independent global protein interaction analyses in yeast using this approach revealed low level of agreement among studies. Can we do better and how?
2. Assessment of the feasibility of a global analysis of the synaptic protein interactome by using a conventional protein-specific immuno-precipitation approach in conjunction with proteomics. This approach requires the availability of low cost-good quality antibodies against all proteins of interest, i.e. it may need a Genome/Organelle-wide production of antibodies. Selection of nodes in the network may compromise providing a good network overview. How do we tackle the synaptic interactome starting out with a limited number of proteins in a pull-down? The outcome of this Think Tank may lead to the generation of a new platform for protein-protein interaction studies based on well-discussed standard operation procedures within NISB.
